S-Adenosyl methionine (SAM), also known as AdoMet, is a compound involved in metabolic processes. It serves as a major methyl group donor in a variety of biochemical reactions and is crucial for the synthesis, activation, and metabolism of hormones, proteins, and phospholipids.

Overview[edit | edit source]

AdoMet is synthesized in the body from adenosine triphosphate (ATP) and methionine by the enzyme methionine adenosyltransferase. It is involved in the methylation of DNA, RNA, proteins, and other molecules, a process critical for the regulation of gene expression, protein function, and cell metabolism. AdoMet plays a key role in the synthesis of creatine, epinephrine, and melatonin, and in the metabolism of phospholipids, which are essential components of cell membranes.

AdoMet acts as a substrate in three main types of biochemical reactions:

  • Transmethylation, where a methyl group is transferred to an acceptor molecule.
  • Transsulfuration, where AdoMet contributes to the synthesis of cysteine, glutathione, and other sulfur-containing compounds.
  • Aminopropylation, which is involved in the synthesis of polyamines, compounds that play roles in cell growth and proliferation.

Due to its role in methylation and other biochemical pathways, AdoMet is studied for its potential in treating various health conditions, including depression, liver disease, and osteoarthritis. It is available as a dietary supplement in some countries and is being investigated for its therapeutic effects, though more research is needed to fully understand its benefits and risks.

AdoMet supplementation is thought to increase the availability of methyl groups for methylation reactions, potentially benefiting conditions associated with impaired methylation processes. However, the efficacy and safety of AdoMet supplementation vary, and it should be used under the guidance of a healthcare provider.

Safety and Side Effects[edit | edit source]

While generally considered safe when taken as directed, AdoMet can cause side effects in some individuals, including nausea, diarrhea, and other gastrointestinal symptoms. It may also interact with certain medications, such as antidepressants, and conditions, warranting caution.
