5'-Guanidinonaltrindole (5'-GNTI) is a chemical compound that acts as a selective kappa-opioid receptor (KOR) agonist. It is structurally related to the compound naltrindole, which is a selective delta-opioid receptor (DOR) antagonist. 5'-GNTI has been used in scientific research to study the role of KORs in various physiological and pathological processes.

Chemical Structure and Properties[edit | edit source]

5'-GNTI is a derivative of naltrindole, with a guanidine group attached to the 5' position of the molecule. This modification confers selectivity for the kappa-opioid receptor over other opioid receptors such as the mu-opioid receptor (MOR) and the delta-opioid receptor.

Pharmacology[edit | edit source]

As a selective kappa-opioid receptor agonist, 5'-GNTI binds to and activates KORs, which are a type of G protein-coupled receptor (GPCR). Activation of KORs by 5'-GNTI can produce a range of effects, including analgesia, dysphoria, and diuresis. KORs are also implicated in the regulation of mood and stress responses, making 5'-GNTI a valuable tool for studying these processes.

Research Applications[edit | edit source]

5'-GNTI has been utilized in various research studies to investigate the role of kappa-opioid receptors in pain modulation, addiction, and mood disorders. For example, it has been used to explore the potential therapeutic effects of KOR agonists in treating conditions such as depression, anxiety, and substance use disorders.

Side Effects and Limitations[edit | edit source]

While 5'-GNTI is a useful research tool, its activation of kappa-opioid receptors can also produce undesirable side effects such as dysphoria and hallucinations. These effects limit its potential for therapeutic use in humans.
