Allelic Heterogeneity refers to the phenomenon where different mutations at the same locus cause a similar or identical phenotype. Loss-of-function mutations result in a gene product with reduced or no function. These can occur through various mechanisms, including nonsense mutations, which introduce a premature stop codon, and frameshift mutations, which alter the reading frame of the gene.

Gain-of-function mutations, on the other hand, result in a gene product with enhanced, new, or detrimental function. These can occur through mechanisms such as missense mutations, which change a single amino acid in the protein product, and splice site mutations, which can alter the splicing of the gene transcript.
