Neutrophil Collagenase

Neutrophil collagenase, also known as Matrix metalloproteinase-8 (MMP-8), is an enzyme that is primarily produced by neutrophil granulocytes. It is part of the matrix metalloproteinase family, which are proteolytic enzymes responsible for the degradation of extracellular matrix components.

Function[edit | edit source]

Neutrophil collagenase plays a crucial role in the breakdown of the extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. It specifically targets type I, II, and III collagens, which are the main structural components of many tissues, including cartilage, bone, and skin.

Structure[edit | edit source]

Like all matrix metalloproteinases, neutrophil collagenase is composed of several domains: a propeptide domain, a catalytic domain, a hinge region, and a hemopexin-like domain. The enzyme is initially produced as a latent proenzyme and is activated when the propeptide domain is removed.

Clinical significance[edit | edit source]

Overexpression of neutrophil collagenase has been implicated in several pathological conditions, including rheumatoid arthritis, periodontitis, and various types of cancer. In rheumatoid arthritis, the enzyme contributes to the destruction of cartilage and bone. In periodontitis, it is involved in the breakdown of periodontal tissue. In cancer, it can facilitate the invasion and metastasis of tumor cells by degrading the extracellular matrix.

Inhibition[edit | edit source]

Given its role in various diseases, neutrophil collagenase is a potential target for therapeutic intervention. Several types of MMP inhibitors have been developed, but their clinical use has been limited due to side effects and lack of specificity.
