Succinyl-CoA synthetase (SCS), also known as succinate-CoA ligase or succinyl-CoA ligase, is an enzyme that plays a crucial role in the metabolism of cells, particularly within the Krebs cycle (also known as the citric acid cycle or TCA cycle). This enzyme is responsible for the conversion of succinyl-CoA to succinate, a reaction that is unique as it couples the hydrolysis of adenosine triphosphate (ATP) or guanosine triphosphate (GTP) to the formation of succinate and Coenzyme A (CoA).

Succinyl-CoA synthetase catalyzes the following reversible reaction:

Succinyl-CoA + Pi + ADP ↔ Succinate + CoA + ATP

In some organisms, the enzyme operates in the reverse direction, synthesizing succinyl-CoA from succinate as part of the glyoxylate cycle. This enzyme is a key player in the energy production processes of cells, linking carbohydrate, fat, and protein metabolism with the production of ATP, the energy currency of the cell.

Succinyl-CoA synthetase is a heterodimer composed of an α-subunit and a β-subunit, which together form the active site of the enzyme. The structure of the enzyme allows for the binding of succinyl-CoA and the nucleotide substrates (ADP or GDP) for the enzymatic reaction. The enzyme's structure and function can vary slightly between different species, reflecting the enzyme's adaptation to different metabolic requirements.

The mechanism of action of succinyl-CoA synthetase involves the formation of a phosphorylated enzyme intermediate. This intermediate then reacts with ADP or GDP, forming ATP or GTP and releasing succinate and CoA. This reaction is an example of substrate-level phosphorylation, a process by which ATP is formed directly in the Krebs cycle.
